Overall Goal of Core B:
Core B will provide support for all projects with novel knockout or transgenic mice with endothelial gene alterations and in vivo metabolic measurements of metabolic phenotype.
Core B will develop verified, internally consistent colonies of experimental models including novel mice with endothelial specific modulation of relevant target genes. In collaboration with Dr. Lin Gan (consultant), Director of our Transgenic & Genomic Editing Core, Core B we have created cutting edge CRISPR/Cas9-generated floxed knock-in mutant mice that can reduce or enhance glycolysis (Glyco-Lo or Glyco-Hi) and these mouse models will be maintained in Core B and will be shared with all 3 Projects. Core B will also provide the latest high resolution techniques to assess cardiometabolic phenotyping of experimental animals including Comprehensive Lab Animal Monitoring System (CLAMS) for whole body metabolism and activity, NMR for body composition and markers of metabolic status in the blood and urine.

Dr. David Stepp
Being a Leon Henri Charbonnier Professor of Physiology in the Vascular Biology Center at the Medical College of Georgia at Augusta University, Dr. Stepp is a leading expert on the cardiovascular consequences of obesity with over 30 years’ experience in conducting animal research in a number of model species and over 10 years’ experience in generating novel mouse models of metabolic vascular disease such as obesity and diabetes.
This includes expertise in breeding and phenotyping mice with an in depth focus on metabolic and cardiovascular phenotypes relevant to this program. His lab is appropriately staffed and equipped to maintain all the colonies used in the program across all projects and to perform the phenotypic characterization of novel mice.
